Identification of ‘Candidatus Phytoplasma’ species in “huanglongbing” infected citrus orchards in the Caribbean

چکیده
Abstract “Huanglongbing” (HLB) is one of the most devastating diseases citrus orchards worldwide. Samples from 183 plants different cultivars and rootstock/cultivar combinations, showing HLB symptoms in three Caribbean countries (Cuba, Jamaica, Guadeloupe-France), were collected to verify possible co-infection ‘ Candidatus Phytoplasma’ Liberibacter’ species. The 64% samples resulted positive Ca . L. asiaticus ’ 27% diverse Phytoplasma’-related species, moreover about 14% infected with also found phytoplasmas, indicating presence mixed infection especially located Cuba. Moreover, Jamaica phytoplasma was detected. Moreover detection only phytoplasmas 11 symptomatic Cuba Guadeloupe without detection, confirmed that symptomatology cannot be sole criterium discriminate between two pathogens, molecular necessary identify single or infections. Diaphorina citri insects asiaticus’ confirming its active role dissemination pathogen. Only insect Cicadidae family, Guadeloupe, for presence. Considering belonging some species’ detected varieties, a relevant as reservoir can attribute orchards.

16S/23S intergenic spacer regions from the rRNA operons of two strains of “Candidatus Liberobacter asiaticum” and one strain of “Candidatus Liberobacter africanum” were cloned and sequenced. The intergenic spacers of the two “Candidatus L. asiaticum” strains studied are identical and contain the genes for isoleucine tRNA (tRNA”‘) and alanine tRNA (tRNAMa) separated by 11 nucleotides. The interg...

Citrus huanglongbing (HLB, ex greening) is one of the most serious diseases of citrus. Different forms of the disease are caused by different Candidatus Liberobacter species, Candidatus Liberibacter asiaticus (Las), Ca. L. africanus (Laf) and Ca. L. americanus (Lam). The pathogen is transmitted by psyllid insects and by budding with contaminated plant materials. The vector psyllid Diaphorina ci...

Leafhoppers (Hemiptera: Auchenorrhyncha: Cicadellidae) account for more than 80% of all "Auchenorrhynchous" vectors that transmit phytoplasmas. The leafhopper populations in two almond witches'-broom phytoplasma (AlmWB) infected sites: Tanboureet (south of Lebanon) and Bourj El Yahoudieh (north of Lebanon) were surveyed using yellow sticky traps. The survey revealed that the most abundant speci...

Citrus huanglongbing (HLB), also known as citrus greening (3), is considered the most serious disease of citrus worldwide (8). The disease has long been present in Asia, Africa, the Indian subcontinent, the Mascarene Islands, and the Arabian Peninsula (2). It has only recently been found in South and North America (16,26). The pathogen is a gram-negative bacterium belonging to the alpha subdivi...
